These positions are in our Bioassay, Molecular Biology & Protein Development department within VI Antibody in Tarrytown, NY. There are 5 openings for Summer 2020. Each opening is described below.

Position 1:
This position is with the Therapeutic Proteins - Protein Development Group. The student will learn how to express protein in E. coli, isolate inclusion bodies, refold, & purify fluorophore-labeled complexes which are associated with human T-cells in the immune system. He or she will learn the chromatographic methods & equipment used to purify protein. Additionally, the intern will become familiar with some of the basic concepts & biochemistry of developing immunotherapies.

Position 2:
Antibody drug conjugates (ADCs) represent a rapidly growing therapeutic approach in the field of oncology. A thorough understanding of target and antibody intracellular trafficking remains a key driver in selection of both the target and the antibody in order to selectively deliver the cytotoxic payload. We developed a new method to simultaneously measure antibody binding, internalization, lysosomal localization, and the kinetics of antibody degradation in live cells utilizing a novel FRET-based cathepsisn B dependent cleavable fluorescent biosensor. The intern will apply this new method and develop new methods to quantitatively measure antibody recycling in living cells. First, the candidate will apply biosensor to measure antibody internalization and cleavage kinetics in lysosomes. Second, the intern will develop a fluorescence based method to quantitatively measure how much payload antibody can delivery for each cell. Finally, the intern will apply the combine method to a lot of targets in many tumor cell lines. Th intern will become familiar with genernal cell biology concepts, cell culture, cell imaging, high content screening, cell based assay development, and antibody interanlization and trafficking.

Position 3:
This internship will focus on the development of cell-based assays to measure the biological activity of antibodies and to support screening and lead candidate selection. This will involve mammalian cell culture, luciferase assays, flow cytometry, and confocal microscopy.

Position 4:
This position is in the Therapeutic Proteins department. The student will be part of a fast-paced bioassays group involved in cutting-edge work to develop novel therapeutics. This student will primarily focus on cell line engineering and cell-based assay development for ongoing therapeutics programs. He/she will also get the opportunity to learn about high-throughput screening using cell based assays. Good communication skills and passion for science are essential.

Position 5:
The intern will work in the department of Therapeutic Proteins, in the Bioassay, Molecular Biology & Protein Development group. The focus will be the production and characterization of amyloidogenic, fibrillizing protein reagents which are involved in neurodegenerative 'proteinopathies', which include proteins such as Tau, ?-synculcein, amyloid-?, TTR, & TDP-43 proteins. The intern will develop methods to form and isolate various aggregated states (monomer, oligomer, fibrils, etc.), which may be present in human samples or conditioned media. They will investigate parameters which affect the misfolding of fibrillizing proteins, and generate methods to isolate and study these proteins for research programs.

Must be pursuing a Bachelors, Masters, or PhD degree

Majoring in Biology, Molecular Biology, Biochemistry, Biosciences, Microbiology, or Bioengineering

1 year experience in science laboratory

Familiarity with methodology & basic concepts of protein expression & purification

Basic pippetting skills, mammalian cell culture technique, flow cytometry, cell-based bioassay development

Position 5- 1-2 years lab research experience. Buffer preparation, SDS-PAGE, Western blotting, absorbance spectroscopy and microscopy.Knowledge of protein expression in E.coli, and chromatography methods such as affinity, ion exchange, size exclusion, & reverse phase. Good Lab practices, attention to detail, including note taking and understanding of laboratory safety are critical.

General Intern Program Information:

Must be enrolled in, or accepted to, an academic program pursuing a Bachelor's, Master's, PhD or PharmD

Summer Program is full-time (~40 hrs/wk) for at least 10 weeks and is paid

Prefer demonstrated leadership in areas such as campus activities, clubs, sports or the community

Minimum GPA of 3.0

You will work with a specific manager on a project(s)
